The Dutch government wants to reduce the release (leaching) of nutrients from greenhouse horticultural firms with substrate-grown crops into the environment. These releases must be practically nil in 2027. This study examined which measures could be taken by the firms to comply with this objective and the associated costs

    Zuivering van lozingswater (afvalwater) is betaalbaar voor 30-35% van de glastuinbouwbedrijven met substraatteelt (potgrond, steenwol). Deze bedrijven exploiteren 40-50% van het gehele areaal glastuinbouw met substraatteelt. Bij de beoordeling van de betaalbaarheid zijn de extra jaarkosten uitgedrukt als percentage van het bedrijfssaldo en van het inkomen uit bedrijf. Daarnaast is gekeken of de benodigde investeringen kunnen worden gefinancierd uit de vrije investeringsruimte

    Fitting the curve in Excel®:Systematic curve fitting of laboratory and remotely sensed planetary spectra

    Spectroscopy in planetary science often provides the only information regarding the compositional and mineralogical make up of planetary surfaces. The methods employed when curve fitting and modelling spectra can be confusing and difficult to visualize and comprehend. Researchers who are new to working with spectra may find inadequate help or documentation in the scientific literature or in the software packages available for curve fitting. This problem also extends to the parameterization of spectra and the dissemination of derived metrics. Often, when derived metrics are reported, such as band centres, the discussion of exactly how the metrics were derived, or if there was any systematic curve fitting performed, is not included. Herein we provide both recommendations and methods for curve fitting and explanations of the terms and methods used. Techniques to curve fit spectral data of various types are demonstrated using simple-to-understand mathematics and equations written to be used in Microsoft Excel® software, free of macros, in a cut-and-paste fashion that allows one to curve fit spectra in a reasonably user-friendly manner. The procedures use empirical curve fitting, include visualizations, and ameliorates many of the unknowns one may encounter when using black-box commercial software. The provided framework is a comprehensive record of the curve fitting parameters used, the derived metrics, and is intended to be an example of a format for dissemination when curve fitting data

    Archaeorhizomycetes Spatial Distribution in Soils Along Wide Elevational and Environmental Gradients Reveal Co-abundance Patterns With Other Fungal Saprobes and Potential Weathering Capacities.

    Archaeorhizomycetes, a widespread fungal class with a dominant presence in many soil environments, contains cryptic filamentous species forming plant-root associations whose role in terrestrial ecosystems remains unclear. Here, we apply a correlative approach to identify the abiotic and biotic environmental variables shaping the distribution of this fungal group. We used a DNA sequencing dataset containing Archaeorhizomycetes sequences and environmental variables from 103 sites, obtained through a random-stratified sampling in the Western Swiss Alps along a wide elevation gradient (>2,500 m). We observed that the relative abundance of Archaeorhizomycetes follows a "humped-shaped" curve. Fitted linear and quadratic generalized linear models revealed that both climatic (minimum temperature, precipitation sum, growing degree-days) and edaphic (carbon, hydrogen, organic carbon, aluminum oxide, and phyllosilicates) factors contribute to explaining the variation in Archaeorhizomycetes abundance. Furthermore, a network inference topology described significant co-abundance patterns between Archaeorhizomycetes and other saprotrophic and ectomycorrhizal fungal taxa. Overall, our results provide strong support to the hypothesis that Archaeorhizomycetes in this area have clear ecological requirements along wide, elevation-driven abiotic and biotic gradients. Additionally, correlations to soil redox parameters, particularly with phyllosilicates minerals, suggest Archaeorhizomycetes might be implied in biological rock weathering. Such soil taxa-environment studies along wide gradients are thus a useful complement to latitudinal field observations and culture-based approaches to uncover the ecological roles of cryptic soil organisms

    Calsequestrin as a risk factor in Graves’ hyperthyroidism and Graves’ ophthalmopathy patients

    Background: The pathogenesis of Graves’ ophthalmopathy (GO), Graves’ hyperthyroidism (GH) and the mechanisms for its link to thyroid autoimmunity are poorly understood. Our research focuses on the role of the skeletal muscle calcium binding protein calsequestrin (CASQ1) in thyroid. We measured the concentration of the CASQ1 protein correlating levels with parameters of the eye signs, CASQ1 antibody levels and CASQ1 gene polymorphism rs3838284. Methods: CASQ1 protein was measured by quantitative Western Blotting. The protein concentrations were expressed as pmol/mg total protein by reference to CASQ1 standards. Results: Western blot analysis showed the presence of two forms of CASQ1 in the thyroid. The mean concentration of CASQ1 protein was significantly reduced in patients with Graves’ disease, compared to thyroid from control subjects with multi-nodular goitre or thyroid cancer. Although in patients with GO it was lower than that, compared with patients with GH this difference was not significant. Reduced CASQ1 in Graves’ thyroid correlated with the homozygous genotype of the rs3838284 CASQ1 polymorphism. Conclusions: Decreased CASQ1 in the thyroid of patients with Graves’ disease compared to thyroid from control subjects is not explained but may reflect consumption of the protein during an autoimmune reaction against CASQ1 in the thyroid

    Epithelial argininosuccinate synthetase is dispensable for intestinal regeneration and tumorigenesis

    The epithelial signaling pathways involved in damage and regeneration, and neoplastic transformation are known to be similar. We noted upregulation of argininosuccinate synthetase (ASS1) in hyperproliferative intestinal epithelium. Since ASS1 leads to de novo synthesis of arginine, an important amino acid for the growth of intestinal epithelial cells, its upregulation can contribute to epithelial proliferation necessary to be sustained during oncogenic transformation and regeneration. Here we investigated the function of ASS1 in the gut epithelium during tissue regeneration and tumorigenesis, using intestinal epithelial conditional Ass1 knockout mice and organoids, and tissue specimens from colorectal cancer patients. We demonstrate that ASS1 is strongly expressed in the regenerating and Apc-mutated intestinal epithelium. Furthermore, we observe an arrest in amino acid flux of the urea cycle, which leads to an accumulation of intracellular arginine. However, loss of epithelial Ass1 does not lead to a reduction in proliferation or increase in apoptosis in vivo, also in mice fed an arginine-free diet. Epithelial loss of Ass1 seems to be compensated by altered arginine metabolism in other cell types and the liver.Cellular mechanisms in basic and clinical gastroenterology and hepatolog

    Rectal Organoids Enable Personalized Treatment of Cystic Fibrosis

    In vitro drug tests using patient-derived stem cell cultures offer opportunities to individually select efficacious treatments. Here, we provide a study that demonstrates that in vitro drug responses in rectal organoids from individual patients with cystic fibrosis (CF) correlate with changes in two in vivo therapeutic endpoints. We measured individual in vitro efficaciousness using a functional assay in rectum-derived organoids based on forskolininduced swelling and studied the correlation with in vivo effects. The in vitro organoid responses correlated with both change in pulmonary response and change in sweat chloride concentration. Receiver operating characteristic curves indicated good-toexcellent accuracy of the organoid-based test for defining clinical responses. This study indicates that an in vitro assay using stem cell cultures can prospectively select efficacious treatments for patients and suggests that biobanked stem cell resources can be used to tailor individual treatments in a cost-effective and patient-friendly manner
